Pulmonext 5 contains ambrisentan, a specialist medicine for pulmonary arterial hypertension (PAH). What it is used for
Treatment of pulmonary arterial hypertension in appropriately assessed patients, alone or in combination according to specialist guidance. It is not a general treatment for ordinary systemic hypertension.
How it works
Endothelin-1 causes blood-vessel constriction and cell proliferation. Ambrisentan selectively blocks endothelin-A receptors, helping pulmonary arteries relax and reducing pressure load on the right side of the heart.
How to take it
Take exactly as prescribed, with or without food, at a consistent time. Swallow tablets whole; do not split, crush or chew unless the specific product information explicitly permits it. Do not stop independently. Attend all pregnancy tests, blood counts and clinical reviews.
Common side effects
Peripheral oedema, nasal congestion, sinus symptoms, flushing, headache, palpitations, dizziness, tiredness, nausea and reduced haemoglobin may occur. Fluid retention may be more frequent in combination therapy.
When to seek urgent medical help
Contact the specialist urgently for rapid weight gain, worsening swelling or breathlessness, fainting, marked weakness, jaundice, dark urine, persistent nausea, facial or throat swelling or other signs of severe allergy.
Warnings and precautions
Ambrisentan can cause embryo-fetal harm and is contraindicated in pregnancy. Reliable contraception and scheduled pregnancy testing are required for patients who can become pregnant. Tell the specialist about liver disease, anaemia, fluid retention, heart failure, idiopathic pulmonary fibrosis, pregnancy plans or breastfeeding.
Interactions
Cyclosporine can substantially increase ambrisentan exposure and may require dose limitation. The specialist should review all PAH medicines, blood-pressure medicines and other prescription, non-prescription or herbal products.
Missed dose
Follow the patient leaflet or prescriber's instructions. If it is almost time for the next scheduled dose, do not take a double dose to compensate. Ask a pharmacist or prescriber if unsure.
Storage
Keep in the original tightly closed container or blister at room temperature and away from excess moisture. Store out of children's reach and follow the actual pack label.
